The Romero laboratory focuses on the study of the regulation of G protein coupled receptors (GPCR) by intracellular scaffold proteins. We focus primarily on the function and traffic of the parathyroid hormone receptor (PTHR) and the Frizzled family of receptors and their regulation by PDZ proteins.

Problems under investigation include: 1) the modulation of receptor traffic and function by PDZ scaffolds; 2) the role of PDZ proteins in the regulation of the Wnt/Frizzled signaling pathways; and 3) the role of specific PDZ scaffolds in tumor biology.

We apply multiple techniques to these studies, ranging from hard-core biophysical and imaging approaches to studies in whole animals and human patient samples. For example, we apply single-cell and single-molecule fluorescence techniques to the study of the dynamics and traffic of receptors on the plasma membrane. We also use fluorescence correlation spectroscopy approaches to study protein-protein interactions in live cells, confocal and electron microscopy to study the distribution of specific protein targets in subcellular compartments, and other rigorous, quantitative imaging approaches. This work is complemented with protein chemistry, membrane biochemistry and molecular biology studies in normal and transformed cells, and testing of specific signaling models in transgenic animals. The laboratory collaborates seamlessly with the Friedman and Vilardaga labs, sharing equipment, reagents, and research resources.
