"Role of the Tumor Suppressor BRCA2 in Chromosome Damage Repair"

Abstract: BRCA2 is one of several breast tumor suppressors that play crucial roles in DNA damage repair by homologous recombination, a process in which a homologous, undamaged chromatid serves as the information donor in the error-free removal of DNA double-strand breaks and other deleterious chromosomal lesions, such as collapsed DNA replication forks. BRCA2 encodes an exceptionally large protein (≥400 kDa) that harbors functional domains for interactions with DNA and with other DNA repair proteins. Using in vitro reconstitution with highly purified BRCA2 species and companion cell-based and biophysical analyses, our studies have furnished mechanistic insights into the complex DNA damage repair role of BRCA2. Highlights of these studies and their implications will be discussed.
